Simple and Efficient Process for Valorization of Orange Peel Waste for Isolation and Purification of Industrially Important Lipase

Lipase is an industrially important enzyme having a wide range of applications. The present study is focused on the development of a simple and efficient process for isolation and purification of lipase from orange peel waste. The proximate analysis of orange peel waste showed the presence of protein (8.83%) with lipolytic activity (6.11 U/mL). Thus, various commercially available extraction media were tested for the extraction of lipase. Of the tested extraction media, the use of ammonium chloride buffer showed the maximum lipase activity of 16.31 U/mL and specific activity of 231.40 U/mg. The process optimization study was conducted to obtain optimum parameters of extraction from maximum enzyme activity and enzyme specific activity. Under optimized conditions, maximum lipase activity of 49.32 U/mL and specific activity of 594.21 U/mg were obtained. The validation of the process optimization study by using ANNOVA showed the strong correlation between the experimental and predicted values with a significant P-value (< 0.005). Integration of ultrafiltration and adsorption significantly increases the purity by 1.4 fold with enzyme activity of 282.85 U/mL and enzyme specific activity of 890.85 U/mg. The characterization of obtained crude and purified lipase was conducted by using ATR-FTIR and SDS-PAGE. Thus, the present process provides a simple, economical, and industrially scalable approach for valorization of orange peel waste for isolation of the industrially important enzyme lipase.
